PERFIL DE PROTEÍNAS DE LAS SEMILLAS DE “SACHA INCHI” (PLUKENETIA VOLUBILIS L. Y PLUKENETIA HUAYLLABAMBANA BUSSMANN, TÉLLEZ & GLENN)

Resumen

En el Perú Plukenetia es un género de plantas tropicales de la familia Euforbiácea, denominadas inchi, sacha inchi, sacha maní, maní del Inca o maní jíbaro, las cuales se utilizan como alimento de las tribus de la Amazonía desde tiempos ancestrales. Plukenetia volubilis es la especie más conocida, mientras que, Plukenetia huayllabambana es una especie descubierta en la provincia de Rodríguez de Mendoza (Amazonas - Perú) por lo que el objetivo de nuestra investigación fue obtener y cuantificar el perfil de proteínas de las dos especies. Las semillas fueron secadas, molidas y desgrasadas empleando el método de Folch (cloroformo: metanol 2:1), para luego realizar el fraccionamiento de las proteínas aplicando el método de Osborne: albúminas (Agua bidestilada), globulinas (NaCl 10,5 M), gliadinas (Etanol 70%) y glutelinas (NaOH 0,1 M). El análisis químico proximal para cada especie se realizó siguiendo las técnicas propuestas por la AOAC. La cuantificación de proteínas totales fue por el método de Kjeldahl y las proteínas solubles por el método de Gornall (Biuret), siendo las fracciones separadas por SDS-PAGE 15%. Los resultados demuestran que el contenido de proteínas totales en P. volubilis es mayor que P. huayllabambana (30,67 % vs. 27,91 %). Sin embargo, en ésta última el contenido de grasas es mayor (52,11 % vs. 45,49 %). La fracción proteica mayoritaria para P. volubilis fueron las globulinas y para P. huayllabambana fueron las albúminas. Los patrones electroforéticos no muestran diferencias cualitativas, pero sí existen diferencias cuantitativas (p<0.05) entre ambas especies.
